Role of Transient Receptor Potential Melastatin(TRPM) Channels in the Stimulation of Melanin Synthesis by Diazoxide

Abstract

|
The known inhibitors of Ca2+-permeable transient receptor potential melastatin(TRPM) channels, flufenamic acid(FFA) and clotrimazole significantly blocked the diazoxide-induced increase in intracellular Ca2+ concentration([Ca2+]i) and melanin synthesis in B16 melanoma cells. H2O2 which has been shown to stimulate the activity of the TRPM channels, increased [Ca2+]i and melanin synthesis, which was significantly inhibited by FFA. These results suggest that TRPM channels may play an essential role in the diazoxide-induced melanogenesis through [Ca2+]i increase.
